The MPlayer GUI works fine for me under Cygwin.  You can select a file 
with the file selection dialog and play it.
You can specify a file on the command line with DOS format file name.  
Cygwin complains the first time it sees
a DOS format file name, but it still works.  The complaint can be 
disabled.  If you specify the file in DOS format on
the command line in Cygwin's bash shell you have to use double 
backslashes.  If you have all the
proper Cygwin development packages installed, you can even compile a 
MPlayer in which "-vo x11" will work.

I don't know what would be the point of a Wine mplayer.  The GTK version 
works fine.  Wine has never seemed to me
to be more than than an exercise of showing that yes it could be done.  
I used to enjoy playing Freecell under Wine, but
now AisleRiot works just as well.
