if under unix, use /etc/skel to place the contents of your preferred 
$(HOME)/.mplayer/ directory.

With Windows, mplayer searches C:\WINDOWS\mplayer (on a standard 
installation), I've not seen yet any user-specific options, so I don't 
know there is anything needed to be done.
