'yuva444p10le' -> the 'a' in 'yuva' represents alpha.

Check with

     ffmpeg -i bunny.mov -lavfi 
color=s=hd720:c=pink[bg];[bg][0]overlay=shortest=1" test.mov

In test.mov, do you see pink in place of the transparent parts of bunny.mov?
